The role of amino acids in sweat
Research into sweat composition has revealed that eccrine sweat actively concentrates certain amino acids — particularly histidine, glycine, serine, lysine, aspartic acid, and ornithine — at levels far above their plasma concentrations. These High-Demand Amino Acids (HDAA) are lost continuously during cycling, accelerating as duration and intensity increase.
The sweat duct itself plays a role: it uses amino acids and potassium as part of the mechanism to reabsorb sodium and chloride. As amino acid reserves deplete, this reabsorption becomes less efficient — meaning sodium and chloride are lost at faster rates as the ride progresses. This is one reason electrolyte replacement alone often fails to prevent late-ride cramping.
The fatigue connection
Glycine is required to build the porphyrin ring structure of haemoglobin — the molecule that carries oxygen to working muscles. Histidine makes up 8–10% of haemoglobin by amino acid composition and is a precursor to carnosine, which buffers lactic acid in muscle tissue. When these amino acids are depleted through sweat, oxygen transport and buffering capacity are both compromised.
The result is fatigue that arrives earlier than your aerobic fitness would predict — and that doesn't fully resolve with rest alone.
Closing the gap
